Support to prospecte applicants will be provided by the UJA R&I International Projects Office (OFIPI).Requested expertise (and training provided)The main aim of our laboratory is to understand the molecular mechanisms driving cardiovascular development. During recent years we focused on dissecting the role of the homeobox transcription factor Pitx2 during cardiogenesis by generating atrial and ventricular chamber conditional mouse mutants. While cardiac development was not compromised, atrial-chamber specific conditional mutants displayed impaired electrophysiological parameters, reminiscent of atrial fibrillation. Subsequently we discerned that Pitx2
Wnt
microRNAs signaling pathway is key to maintain electrophysiological homeostasis in the developing and adult heart. Over the last years we have centered our attention to dissect the role of non-coding RNAs during cardiogenesis as well as in cardiac pathologies such as atrial fibrillation and dilated cardiomyopathy, including therein microRNAs and lncRNAs. We have identified key microRNAs and lncRNAs that contribute to these pathologies. We are currently generating genetically engineered mouse models to dissect their role in vivo. A second research line in our laboratory is directed to discern the functional role of microRNAs and lncRNAs during epicardial development. Bulk RNA seq analyses provided us with the bases to investigate the role of distinct differentially expressed transcription factors, microRNAs and lncRNAs during epicardial formation as well as in physiopathological processes in which the epicardium plays a key role, such as cardiac regeneration. In this context, we have generated in our laboratory a mouse cryoinjured model to dissect their role during cardiac regeneration. Additionally, we are also dissecting the role of Pitx2 in the epicardial development by generating tissue-specific conditional mouse models. In summary, our main goal is to dissect the role of distinct transcription factors, microRNAs and lncRNAs during heart formation, using different cellular, molecular biology and genetic tools in mice.Share this page
