The focus of this project is on the natural product class of terpenoids, which is the largest and most chemically diverse class of natural products and hold immense potential for biotechnological applications. ModBioTerp will establish enzyme models, that predict and ab-initio tailor the structure and functionality of terpene synthases and unlock the vast potential of terpenoids and lead the way towards sustainable and innovative biotechnological solutions.The aim of the specific thesis project, is to explore the biosynthesis of terpenoids in liverworts and biochemical characterize a range of microbial terpene synthase-like enzymes. Being part of a large network will enable the applicant to obtain knowledge from several laboratories during the project period.The project includes species collection and participation in network training events apart from the biochemical studies in St. Etienne, along with biotechnological challenges to establish transformation of selected liverworts. The project is divided into the following milestones

  • Expression of at least 20 MTPSL from liverworts within Bazzania, Lophocolea, Frunellaria and Porella species in yeast.
  • Participate in the modelling of 5 MTPSL and perform mutations in these together with oloBion.
  • Characterization in vitro and in vivo in yeast and in plants of both original and mutated MTPSLs.

Overall the target is to expand our knowledge of biosynthesis of liverworts terpenoids that are chemically unique when compared to other plants, and biochemically characterize Microbial-Terpene-Synthase-like enzymes across the non-seed plants in order to follow their evolution.We are looking for you that want to perform research within Plant biochemistry and molecular biology with a strong focus on terpenoid biochemistry. At Jean Monnet University, Saint Étienne, France you will be able to break new grounds within Plant Biochemistry and join a research project that go beyond current knowledge within bryophyte terpenoid biochemistry. Thus, we are looking for a candidate with a strong background and a strong desire to obtain a PhD degree from our laboratory. You will be part of the doctoral School EDSIS, UJM.The preferred candidate must have a Master of Science (or engineering) degree in Molecular biology and/or Biochemistry or very similar fields, and preferably worked with plant molecular biology and/or terpenoid biochemistry. The candidate should have some knowledge of bioinformatics. We expect that the PhD candidate have some knowledge of enzyme characterization, cloning techniques, analytical chemistry, thus any knowledge in this is an advantage.The candidate must be able to write and speak English at a scientific level.PROFESSIONAL SKILLS AND KNOWLEDGEThe applicant must possess an MSc degree prior to initiation of the PhD study and be able to take up the position no later than January 1st, 2025.At the time of recruitment, it is a requirement that PhD candidates have not been awarded a doctorate degree and are in the first 4 years (full-time equivalent) of their research careers. Furthermore, at the time of selection by the UJM, researchers must not have resided or carried out their main activity (work, studies, etc.) in either France for more than 12 months in the 3 years immediately prior to their recruitment. Short stays, such as holidays, are not taken into account.SALARY AND APPOINTMENTS TERMSThe appointment will be based on agreement signed with the European Union and follow general employment rules for a French University. The period of employment is 3 years fully financed.WORKPLACEThe main part of the project will be performed at the LBVPAM in St. Etienne, France. The PhD student is expected to perform an external stay for three months at oloBion, Barcelona, Spain and one months at KTH, Stockholm, Sweden.
