is seeking a postdoctoral researcher interested in developing a collaborative project to apply to the Marie Skłodowska-Curie Actions (MSCA) Postdoctoral Fellowship (PF) Call 2024 (call deadline 11 September 2024). The Molecular Motors Manipulation lab led by Dr. B. Ibarra is interested in understanding the dynamic and mechano-chemical processes governing the operation of biological and synthetic systems. Using state-of-the-art optical tweezers techniques we measure and manipulate the real-time kinetics and conformational changes of individual molecules. Our research provides information about the activities of biological and synthetic devices that cannot be obtained with classical bulk methods.Research theme(s) proposedDespite the relevance for human health, the molecular mechanisms of the human mitochondrial DNA (mtDNA) replication remain poorly understood. We lack a clear understanding of the kinetic, mechanistic, and dynamic basis governing the functional coupling of the core factors of the human mtDNA replisome. This knowledge gap precludes the comprehension of the molecular mechanisms behind mtDNA replication and the pathogenesis of associated diseases. To address this knowledge gap, the main objective of this proposal is to determine the molecular mechanisms that govern the operational dynamics of the mtDNA replisome and the effect of pathogenic mutations on these mechanisms. To this end, we propose to exploit the unique properties of single-molecule manipulation and visualization techniques, such as optical tweezers with temperature control and optical tweezers with confocal fluorescence microscopy to:

1. Determine the real-time kinetics of individual replisome factors and in situ reconstituted replisomes.2. Determine the mechano-chemical processes involved in replisome operation.3. Quantify the exchange dynamics and stoichiometry of replisome components during DNA synthesis.4.
