Offer DescriptionCO dehydrogenases (CODH) are metalloenzymes that reversibly oxidize CO into CO2. Recently, our collaborators identified CooS5, the protein the closest to CODH without being one. The aim of the LOCO2 ANR/DFG project that finances this position is to use CooS5 to understand what makes a CO dehydrogenase able to oxidize CO, using an approach in which existing CODHs are changed to ressemble CooS5 and vice versa.
The task of the post-doctoral researcher is to characterize the newly produced enzymes via classical biochemical methods and more importantly via protein film electrochemistry, in order to uncover the the molecular determinants of the CO dehydrogenase activity. Part of the project will be to adapt an in-house electrochemical flow cell to the characterization of CO dehydrogenases to use it.The post-doctoral researcher will characterize the proteins purified in the project using classical biochemical methods and protein film electrochemistry, develop and use the electrochemical flow cell, propose possible variants to purifiy and characterize, and rationalize the observables.The work will take place in the group of Christophe Léger in the Laboratoire de Bionénergétique et Ingénierie des Protéines in Marseille. The group is a world expert in protein film electrochemistry, which they use to study a number of metalloenzymes.
