The relevance of the interplay of staphylocoagulase and staphylokinase in the development of Staphylococcus aureus infections” and “The central role of human hemostatic system in formation of Staphylococcus aureus biofilms”.Special admssion for a grant project

“The central role of human hemostatic system in formation of Staphylococcus aureus biofilms”.The aim of the project is to understand the mechanisms of virulence of Staphylococcus aureus, a bacterium that is one of the main causes of severe infections in humans. In particular, the project focuses on how and for what purpose Staphylococcus takes control of the human coagulation (clotting) and fibrinolysis (clot dissolution) system, especially in the context of lung, endocardium and biofilm infections.An important part of the project will be to identify the role of the coagulation system in the formation of staphylococcal biofilms, to identify the mechanisms controlling the expression of staphylococcal proteins controlling coagulation and fibrinolysis, and possibly to analyze the role of the coagulation system in mouse models (using transgenic mice producing human plasminogen, which can be activated by staphylococcal proteins).The project will use a variety of techniques:

• in vitro (protein isolation from bacteria, coagulation measurements, gene expression measurements using reporter proteins and RT-qPCR, laboratory models of biofilms and abscesses, fluorescence and confocal microscopy);
• ex vivo (isolation and analysis of the behavior of human cells in contact with staphylococcal clots and biofilms);
• in vivo (mouse model of infection in transgenic “humanized” mice).

• very good command of English, spoken and written;
• knowledge of molecular biology, biochemistry, and cell culture techniques (e.g. FACS flow cytometry, ELISA, PCR, gene cloning);
• experience in working with laboratory mice or having the necessary training and permits to do so;
• master’s degree or equivalent obtained in a field of exact and natural sciences.
