Offer DescriptionThe aim of the postdoctoral project is to determine the impact of missense genetic variants identified in the NR2F1 gene, a key regulator of brain development, on the heterogeneity of neurodevelopmental abnormalities described in patients with the disease. The post-doc will seek to understand how rare genetic mutations in NR2F1 affect cortical development and circuit assembly in ‘humanised’ mouse models. The project will compare the pathological contribution of representative human mutations in different mouse models to understand their phenotypic heterogeneity and unveil pathophysiological mechanisms common to multiple neurodevelopmental diseases. The research project will use histological, molecular, and cellular analyses combined with 3D imaging and scRNAseq transcriptomics.– Participate with the research assistant in the organization of transgenic mouse colonies by genotyping
– Assist with intraperitoneal injections and perfusions of adult animals
– Participate in the collection and dissection of post-natal and adult brains
– Undertake thin sections of brains using a cryostat
– Implement and adapt the various immunohistochemistry techniques required to analyze brain structures
– Monitor cell migration using the in utero electroporation technique
– Use animal biology analysis and experimentation equipment
– Use specific imaging software (Image J, AxioVision, etc) and statistical software (GraphPad)
– Analysing and communicating data (drafting laboratory notebooks, presenting results at follow-up meetings)
– Be familiar with the literature
– Possibly supervise a Masters student
– Write a research article in English

– Excellent knowledge of the anatomy and neurogenesis processes of the mouse brain at different ages
– Expertise in mouse genetics, cellular and molecular biology, and microscopy
– Extensive experience in animal experimentation (handling mice, intraperitoneal and subcutaneous injections), dissection, perfusion, and in utero electroporation
– Knowledge of imaging, histology (staining, microscopy) and FACS
– Proficiency in Word, Excel, PowerPoint, Graphpad, and image analysis software (ImageJ, Photoshop, etc).
– Organisational skills, planning, creativity and conscientiousness
– Motivation, perseverance, autonomy and thoroughness
– Able to work independently, but able to adapt and work as part of a team
– Oral and written communication in English (French is not compulsory);
– Authorisation for animal experimentation is desirable.
