Offer DescriptionAntibiotic resistance is an increasing issue that will be a major health challenge in the forthcoming decades. The development of new strategies to fight infections is therefore an urgent necessity, especially against Gram-negative bacteria (GNB). The low permeability of the envelope of these bacteria is the first line of defense against antibiotics. Nutrient assimilation systems are breaches in this bulwark for those who know how to exploit them. The uptake pathways for iron, a crucial element for bacterial proliferation, are particularly promising targets. The majority of bacteria excrete siderophores, iron(III)-chelating metabolites, to chelate this metal ion in the extra medium. The ferric siderophore is then recognized and transported into the bacterium by a specific transmembrane mechanism involving several proteins. The conjugation of an antibiotic with a siderophore leads to hijacking these nutrient assimilation pathways to make antibiotics penetrate the bacteria more efficiently through the Trojan Horse strategy. In the framework of the RHEINGOLD project, we want to vectorize antibacterial gold(I) complexes that are difficult to penetrate into GNBs, using siderophores. The role of the recruited person will be to prepare (synthesis and hemisynthesis), purify and characterize the siderophore vectors, under the direction of Dr. Gaëtan Mislin (HDR University of Strasbourg, DR CNRS, France). The gold(I) complexes will be prepared by our collaborator in Caen (LCMT, ENSICaen, Caen, France) and the conjugation of the two chemical entities will be the subject of close collaboration between the two teams. The siderophore-gold(I) conjugates will be further tested at the National Reference Centre for antibiotic resistance (Besançon, France).The activity of the candidate selected for this project will be mainly focused on organic synthesis. The selected person will also be in charge of the purification and spectroscopic characterization (NMR, MS) of the compounds synthesized. It will nevertheless be possible to be involved in the evaluation of the biological properties of the conjugates. Research is also based on communication: the participation of the recruited person to a congress is budgeted. The writing of articles, and more specifically experimental parts and supporting information will be devoted to the recruited person. Finally, the recruited person will contribute to the technical life of the laboratory alongside other staff members: ordering, maintenance of scientific equipment, management of stocks and reaction waste.The team “Metals and Microorganisms: Biology, Chemistry and Applications” (MMBCA) is member of the UMR7242 Biotechnology and Cell Signaling located at the Ecole Supérieure de Biotechnologie de Strasbourg (ESBS) located at the Illkirch innovation center (10 km from the city center of Strasbourg). This team is composed of chemists and biologists (25 people including 9 permanent positions) developing project at the interface of biology and chemistry with a particular focus on iron homeostasis in pathogenic bacteria. In this context, the research axis dedicated to the delivery of antibacterial organometals is currently a booming field. The team has two chemistry laboratories. Each researcher has for his own use a hood, a rotary evaporator and a diaphragm pump. Each of the two laboratories is equipped with an automated purification system (Chromatoflash). A HPLC, a cryostat (-40°C), a lyophilizator and a microwave reactor are in common access. – Ability to work/evolve in a multidisciplinary environment at the interface of chemistry and biology.
